High-Resolution U/HPLC Separation of Peptides BIOshell™ A160 Peptide Phenyl-Hexyl

High efficiency separation of peptides with orthogonal selectivity to alkyl-bonded phases due to rigid aromatic ring (shape selectivity).

The BIOshell™ A160 Peptide Phenyl-Hexyl bonded phase is based on fused-core particles. BIOshell™ A160 Peptide Phenyl-Hexyl provides a stable, reversed phase packing with a pore structure and pore size that is optimized for reversed-phase HPLC separations of peptides and polypeptides, using typical acidic mobile phases favored for protein structure-function and proteomic applications. The aromaticity of the stationary phase ligand can interact with amino acids with aromatic side chains, allowing for differential selectivity not afforded by alkyl-bonded phases.

Having a phenyl-hexyl bonding, methods developed on this column should be screened with both methanol and acetonitrile, supplemented with an ion-pairing reagent like 0.1% (v/v) trifluoroacetic acid (TFA) or difluoroacetic acid (DFA), as these two solvents have a wider eluotropic strength with stationary phases with aromaticity than is seen with alkyl-bonded phases. For typical peptide workflows, column temperatures of 40 – 60 °C are used; higher temperatures can enhance peak shape and can increase the throughput of the workflow.
